چکیده

the potential changes in neuromuscular blockade after chronic (>24 h) administration of sub-paralytic doses of two newly introduced aminosteroidal muscle relaxants were investigated. org-9426 (rocuronium) and org-9487 were studied in the in vivo guinea pig gastrocnemius muscle-sciatic nerve preparation. the problems seen after prolonged administration of muscle relaxants are thought to arise from up-regulation of achrs and this phenomenon may be responsible, at least in part, for prolonged muscle paralysis, development of resistance to muscle relaxants and generalized muscle weakness seen with chronic administration. after two weeks of chronic treatment, using implanted osmotic mini-pumps delivering sub-paralytic doses of the test drugs, responsiveness to the muscle relaxants altered significantly so that higher doses were required in the drug pretreated groups to produce the same twitch block as in matched control animals. a possible explanation for this observation is that, following chronic treatment, upregulation of achrs occurs, at the neuromuscular junction, thus increasing the required doses in the drug pre-treated animals as compared with the control group.

Nondepolarizing neuromuscular blocking drugs and train-of-four fade.

The aim of this study was to examine differences in prejunctional effects of different relaxants by measuring the train-of-four (TOF) fade during the onset and recovery of neuromuscular block. The relaxants studied were atracurium (225 micrograms.kg-1), mivacurium (65 micrograms.kg-1) rocuronium (300 micrograms.kg-1) and vecuronium (40 micrograms.kg-1). The TOF ratios were measured at approxima...

Neuromuscular Blocking Agents

• Neuromuscular blocking agents are used to improve conditions for tracheal intubation, to provide immobility during surgery, and to facilitate mechanical ventilation. • The main site of action of neuromuscular blocking agents (muscle relaxants) is on the nicotinic cholinergic receptor at the endplate of muscle. They also have effects at presynaptic receptors located on the nerve terminal. • Su...

Effects of prostaglandins and indomethacin on neuromuscular blocking agents.

The effects on neuromuscular blockade by d-tubocurarine and succinylcholine of inhibition of prostaglandin biosynthesis by indomethacin and of intra-arterial administration of prostoglandins E2 and F2 alpha, before and after inhibition of prostoglandin biosynthesis, were evaluated in the cat sciatic-tibialis preparation. Non-specific inhibition of prostaglandin biosynthesis by indomethacin 3 mg...

Neuromuscular blocking agents: a review.

The author provides a review of the mechanism of neuromuscular transnission, as well as the Nechanisn of action and differences between curare and succinylcholine. Effects of these two relaxants on various organs and tissues are discussed, as are their onset of action, duration, elimination, and clinical uses. In order to understand how skeletal muscle re-laxants act, the mechanism of neuromusc...
